Pull theadapter back into theon-column
injector.
Tighten the nut and ferrule securely so
that the columnwill not move even if you
pull on it with your fingers. Donot over-
tighten the ferrule un-necessarily. You
want to feel the ferrule compress a little
bit but not tighten somuch that the fer-
rulebecomesdistorted.
Replace the septumnut. Be careful not to
over-tighten the septumnut. If the septum is
too tightly compressed, the syringeneedle
willmakeahole in the septum. If the sep-
tum is lightly compressed, the silicone sep-
tum cleavesand re-sealspermittinghun-
dreds of injections before failureandwill not
clog the syringeor columnwith septumpar-
ticles.
Insert anempty syringe through the sep-
tumandonto the column to verify that
the syringe slides smoothly into the col-
umnwithout catchingor snagging. If it
feels like the syringeneedle catches on
theendof the column, then reposition
the column1 cm closer to the center of
theoven.
