16. Injecting aSample andStarting theRun
TheRheodyne injectionvalvecomeswith itsownblunt-tippedflushingneedle,
which isshipped inserted into thevalveunder theprotectiveredcap. Remove the
protective cap and tape, thenpull out theneedle. Theprovided100µLglass
injectionsyringeshouldsuitmostinjectionapplications. Forthebestresults,prepare
your sample in thesamesolventyouareusing foryourmobilephase. Fill the
syringewithsample, andeliminateanyairbubbles. Insert thesyringe into the
Rheodyneinjectionvalve,pushinggentlyuntiltheneedlehitsthestop. Depressthe
syringeplungertofillthe20µLsampleloop. Leavethesyringeinthewhileyouturn
the Rheodyne injection valve knob to the INJECT position. The run will
automaticallystart. See theRheodynedocumentation inyourSRImanualunder
HPLCInjectorsformore informationaboutoperating the injectionvalve.

solvent recycle valve may be disconnected, and
connected toexternaldetectors, likerefractive indexor
fluoresencedetectors.Theexternaldetector’sexittubes

RelayCcontrols thesolventrecyclevalve. WhenRelayC
isON, thesolvent isdirected towaste (greenexit tube).
TurnRelayCONbeforeanypeaksappearonscreen,and
turn itOFFat theendof the run. ClickonRelayC in the
Relays/pumpwindow to turn itONandOFF, or usean
eventtabletodoitautomatically. Thesolventrecyclevalve
may alsobeused as a singlepeak fraction collector by
turning itON/OFFatspecific timesduring therun.
