14. Priming thePump
Note:
Donotopen theprime/purgevalveuntilpressure is
atzero,orveryclose.Otherwise, systempressurecould
damage thepulsedamperorcause thecolumnadsorbent
bed toshift.
Open theprime/purgevalveby turning it to the left, then
startthepumpbyclickingonRelayAinthepumpwindow.
Inserttheprimingsyringeintotheprime/purgevalve, then
slowlyandgentlypullbacktheplunger.Whenyoucanpull
abubble-lessstreamoutof theprime/purgevalve, close
thevalveby turning itback to the right.Thepressureshouldbeginbuilding tooperating range,which is
dependentupon the flow rate, solventviscosity, and thebackpressureof thecolumnselected.When the
systemstabilizesat theoperatingpressure range,youmay injectyoursample. It isnormal for thepressure
gaugeneedle tofluctuatewith thepumpstroke.

. In theRelays/pumpwindow, clickonRelayAto turn the
pumpOFF,sinceyouhaven’tprimed ityet. Theflowrateof thepump(pumpAinabinarygradientsystem)
canbeadjustedfrom0.1 to5.0mL/minute, in incrementsof0.1mL/minute,using thedialogboxat thebottom
oftheRelays/pumpwindow. Inabinarygradientsystem,thegradientprogramdetermineshowtheflowissplit
between the twopumps.
