Fattyacidmethylesters (FAMEs)
FAMEs analysis is important in fats andoils charac-
terization and in the determinationof total fat con-
tent in foods. To prepare themethyl esters, fats are
extracted frommatrices using non-polar solvents,
and saponified to produce free fatty acids. After
derivatization to the methyl esters, molecules have
increased volatility and decreased activity, which
permitsmore accuratequantitationbyGC.
Capillary columns withpolyethylene glycol (PEG)
or Carbowax® stationary phases are used to ana-
lyze saturatedandunsaturatedFAMEs.For the res-
olution of the
cis
and
trans
isomers, bis-
cyanopropyl phases typicallyareused.
Stabilwax®
and
Rtx®-Wax
columns provide excellent resolu-
tionof FAMEsderived frombothplant andanimal
sources.
FAMEWAX®
columns offer excellent reso-
lutionof polyunsaturatedFAMEswith significant-
ly reduced analyses times, compared to traditional
Carbowax® stationary phases. Individual
cis
and
trans
isomers are resolvedon an
Rt-2560
column,
making it the column of choice for analyzing par-
tiallyhydrogenated fats.
