How toAvoidPressureTransients
Air in the sample loopcancausean instantaneous systempressure
drop thateventually returns toanormal level.Air causes thepressure
todropwhen the injectormoves from theLOAD to the INJECT
position.When large sample loops (≥100µL) arepartially loaded, air
present in theneedleport tube ispushed into the sample loop (see
Figure1).Air canalsoenter the sample loop from siphoningwhich
occurswhen thevent line ishigher than the injectionport. Ineither
case, upon injection, the systempressurecollapses theairbubble,
causingpressure todropmomentarily.
Apressuredrop in the systemcausedbyair results inchanges in
retention time, artifact peaks, andaffects columnperformance.
Avoidpressuredropsby removing theair in theneedleport tube.
Do thisbyflushingabout 1mLofmobilephasewitha luer syringewith
needleport cleaner. Keep theneedleport tubefilledwithmobilephase
byoccasional flushing.Adjust thevent line(s) so theoutlet isat the
samehorizontal level as theneedleport (seeFigure2). For additional
