If thecolumnbed shrinks adeadvolumecaneasilybe removedbymovingdown thevariable
piston (see page 4, chapter 1).
CAUTION! -Only use degassed and pre filtered solvents for operating the column.
Particlesmay clog the frits or damage the column packing!Make sure that the particle size of
the chosen chromatography packing correlates to the frit porosity of the column!
Hints for operation:
5.1Storage of the packed column:Open the sealing stoppers for one turn allowing the
compensationof temperature-dependent pressure changes.
CAUTION! - Protectmoistened column against intensive heat and direct insulation. The
heat induces the evaporation of high-volatile solvents, and the resulting pressure can crack
the column.
5.2We recommend tooperate the column from the bottom (variable endpiece) to the top (fixed
endpiece).The resultingbenefit is that air escapes faster from the column so that it is
conditioned faster (i.e.with less solvent).
5.3We recommend tomake surebefore sample application that nodeadvolumeoriginated at the
column inlet during theconditioningphase (eliminationof deadvolume seepage6, chapter 4).
6.0 SOLVENTRESISTANCE:
Packed columns canbe stored either in20/80 ethanol/water, containingup to1MNaCl and
neutral buffer or inneutral buffer containing0,03% sodiumazide.NPandRPcolumnsmaybe
stored inorganic solvents as needed for commonuse.All solvents shouldbe filtered through a
0.45um filtermembrane.
Ingeneral the following solvents and additives canbeused innormal operation.Howeverwedo
not recommend touse anyof them for longer periods of time or for storage. For information
please refer toEssentialLifeSolutionsLtd.
