ChoosingaStationaryPhase
Identifying the appropriate stationaryphase for your separation is themost critical stepof column selec-
tion. The choice of stationary phase should be based on sample solubility and on chemical differences
among the sample compounds that canbe exploited to separate the analytes of interest (Figure 1).
Sample hydrophobicity also is amajor determinant of the separationmode (reversed phase or normal
phase). In reversedphase separations, themobile phase ismorepolar than the stationaryphase,which is
traditionally a straight alkyl chain,most oftenoctadecylsilyl,C18 (ODS).ThemajorityofHPLC analyses
are performed in reversedphasemode becausemost analytes of interest canbe dissolved inmixtures of
water and/or apolar organic solvent such asmethanol or acetonitrile.
Innormal phase separations, themobilephase is less polar than the stationaryphase (e.g.,whenbare sil-
ica is the stationary phase and hexane with amodifier is themobile phase). The normal phasemode is
usedprimarilywhen the reversedphasemode cannot beused, because reversedphase separations gener-
ally aremore robust, reproducible, and versatile. Note that a stationary phase incorporating both polar
and nonpolar functionality can be used in either reversed phase or normal phasemode. Cyano phases
(e.g.,Ultra, Pinnacle™DB, or Pinnacle™ II cyano columns) commonly areused in either reversedphase
or normal phasemode.Ultra IBD andAllure™Basix columns also canbeused for either reversedphase
ornormal phase separations.Allure™PFPPropyl andUltraPFP columns alsodisplaydual functionality.
