Dissolved oxygen can cause flow rate instability and increased baseline
noise.Also, ithas aquenchingeffecton fluorescencedetectionand increas-
es the background of UV detectors. Dissolved gases can out-gas in the
HPLC system, formingbubbles incheckvalves, at connections,or indetec-
tor flow cells.
In-line vacuumdegassing ismore effective at removingdissolved gas from
mobile phases than sonication or helium sparging.Most in-line degassers withdraw gas across a gas-
permeable PTFE tubingmembrane, but the Degasys Ultimate Degasser uses tubing composed of an
amorphous fluoropolymer that is 200 to300 timesmoregaspermeable thanPTFE.This translates into
the ability to use shorter tubing for removing dissolved gas. This newmaterial also has better tubular
burst strength thanPTFE. To prevent cross contamination, each channel on this Degasys unit is indi-
vidually encasedwithin its own vacuum chamber.
Not recommended for usewith fluorinatedmobile phases, includingmobile phases containingTFA.
